石棉致细胞毒性的氧化应激机制的研究进展

Research progress on oxidative stress related mechanism of asbestos cytotoxicity

  • 摘要: 石棉与石棉肺、胸膜斑、肺癌和恶性间皮瘤等疾病密切相关,其致病机制尚未完全阐明。石棉纤维可诱导活性氧(ROS)的产生,ROS的堆积或抗氧化物的缺乏均可使机体内氧化-抗氧化系统失衡,导致细胞发生脂质过氧化、蛋白质变性、DNA损伤和酶活性改变等一系列的氧化应激损伤,是石棉的致病机制之一。对ROS的生成及其致机体氧化应激损伤分子机制的深入研究,可能为石棉暴露相关性疾病的预防、诊断和治疗提供思路。

     

    Abstract: Asbestos is closely associated with asbestosis, pleural plaques, lung cancer, and malignant mesothelioma; however, their pathogenic mechanisms have not been fully elucidated. The accumulation of reactive oxygen species induced by asbestos fibers or the lack of antioxidants can cause oxidation-antioxidant system imbalance, leading to a series of oxidative stress injuries, including lipid peroxidation, protein denaturation, DNA damage, and enzyme activity alterations, which is one of the potential pathogenesis of asbestos. Further studying the production of reactive oxygen species and the molecular mechanism of oxidative stress injury may provide new ideas for the prevention, diagnosis, and treatment of asbestos exposure related diseases.
