  • 摘要: 目的 探讨热休克蛋白HSP70-1+190(G/C)、HSP70-2+1267(A/G)两位点基因多态性与肺癌易感性的相关性。

    方法 采用病例-对照研究,以病理确诊的肺癌患者159例为病例组, 202名正常体检人群为对照组。应用聚合酶链反应-限制性片段长度多态性分析(PCR-RFLP)技术检测HSP70-1+190(G/C)和HSP70-2+1267(A/G)的单核苷酸多态性。采用非条件Logistic回归分析两位点基因型与吸烟的交互作用及两位点之间的交互作用。

    结果 HSP70-1+190(G/C)位点的3种基因型GG、GCCCGC等位基因频率在病例组和对照组之间的分布,差异均有统计学意义(χ2=19.16, P < 0.05; χ2=18.76, P < 0.05), CC基因型与肺癌具有中等强度关联。HSP70-2+1267(A/G)位点的基因型及等位基因的分布在两组之间差异均无统计学意义(P > 0.05)。按照吸烟情况进行分层分析发现,重度吸烟人群中携带HSP70-1+190(G/C)位点的GC基因型和GC+CC基因型者及HSP70-2+1267(A/G)位点的AA基因型的个体患肺癌的风险增高。基因型与吸烟的交互作用分析发现, HSP70-1+190(G/C)位点多态性和吸烟具有相乘交互作用(P=0.01)。HSP70-1+190(G/C)和HSP70-2+1267(A/G)两位点多态性之间存在交互作用(χ2=12.87, P=.00)。

    结论 HSP70-1+190(G/C)位点多态性可能与河南汉族人群中肺癌易感性有关,与HSP70-2+1267(A/G)和吸烟均具有相乘交互作用;HSP70-2+1267(A/G)位点多态性与肺癌的相关性还有待进一步研究,其AA基因型可能与重度吸烟之间存在交互作用。

     

    Abstract: Objective To investigate the association between the heat shock protein 70 gene polymorphism and lung cancer.

    Methods A case-control study was conducted among 159 lung cancer patients and 202 control subjects. Polymerase chain reaction-restrained fragment length polymorphism technique (PCR-RFLP) was applied to detect the single nucleotide polymorphisms (SNPs) of HSP70-1 +190 (G/C) and HSP70-2 +1267 (A/G). The interactions of the two gene polymorphisms and smoking and that of the two sites were analyzed by non-conditional Logistic regression.

    Results There were significant differences between the case group and control group (χ2=19.16, P < 0.05; χ2=18.76, P < 0.05)in the distribution of each genotypes GG, GC and CC, and also in the frequencies of the two alleles G and C of HSP70-1 +190 (G/C). The CC genotype was moderately correlated with lung cancer. There were no significant differences in the distribution of genotypes and allele frequencies of HSP70-2 +1267 (A/G) between the case group and control group (P > 0.05). After stratification analysis according to smoking, it revealed that in heavysmokers subgroups who carried HSP70-1 +190 (G/C)GC or GC+CC genotypes and/or HSP70-2 +1267 (A/G)AA genotype were associated with significantly increased risk of lung cancer. The interaction analysis of genotype and smoking revealed that there was multiplication interaction between HSP70-1 +190 (G/C) polymorphism and smoking on susceptibility to lung cancer. There was also interaction between the two genotypes HSP70-1 +190 (G/C)and HSP70-2 +1267 (A/G)(χ2=12.87, P=.00).

    Conclusion HSP70-1 +190 (G/C)polymorphism may be associated with the susceptibility to lung cancer in He'nan Han population, and there are multiplication interaction with HSP70-2 +1267 (A/G)polymorphism and smoking. The relationship between HSP70-2 +1267 (A/G) polymorphism and lung cancer has yet to be further studied and there may be an interaction between AA genotype and the heavy smokers.
