氰戊菊酯宫内暴露对雄性仔鼠雄性特征的影响

Effects of Intrauterine Fenvalerate Exposure on the Male Characteristics of Offspring Rats

  • 摘要: 目的 了解氰戊菊酯宫内暴露对雄性仔鼠雄性特征的影响。

    方法 选用 SD妊娠大鼠 20只, 随机分为4组, 分别给予 0、2、10、50 mg/kg氰戊菊酯, 于妊娠第 12~18天进行灌胃染毒, 仔鼠出生后第 13天每组随机抽取 35只雄性仔鼠观察乳头保留情况。仔鼠断乳后进行窝标准化, 每组随机保留 25只雄性仔鼠, 并于出生第 35天处死取材, 制备睾丸匀浆, 应用比色法测定睾丸组织一氧化氮(nitric oxide, NO)及一氧化氮合酶(nitric oxide synthase, NOS)水平。

    结果 雄性仔鼠出生第 13天, 2 mg/kg和 10 mg/kg染毒组与空白对照组比较, 乳头保留数差异无统计学意义(P > 0.05), 而50 mg/kg染毒组乳头保留数明显高于空白对照组, 差异有统计学意义(P < 0.05)。与空白对照组相比, 2 mg/kg染毒组 NOS活性无明显改变(P > 0.05), 10 mg/kg和 50 mg/kg染毒组 NOS活性明显升高, 差异有统计学意义(P < 0.05)。各染毒组 NO含量与空白对照组相比, 差异均无统计学意义(P > 0.05)。

    结论 氰戊菊酯宫内暴露对雄性仔鼠雄性特征有明显的类雌激素效应。

     

    Abstract: Objective To investigate the effect of intrauterine fenvalerate exposure on the male characteristics of offspring rats.

    Methods Twenty pregnant SD rats were randomly divided into 4 groups, and intragastrically administrated with 0, 2, 10 and 50 mg/kg of fenvalerate respectively from gestation day 12 to 18. The number of nipple retention was observed of 35 male offspring rats randomly selected from each group on postnatal day 13. And 25 male offspring rats were randomly kept after ablactation in each group and killed on postnatal day 35 to prepare testicular homogenate. The level of nitric oxide (NO) and nitric oxide synthase (NOS) in testicles were measured.

    Results Compared with the control group, the number of retained nipples was significantly increased in 50 mg/kg group (P < 0.05), but not in 2 and 10 mg/kg groups (P > 0.05); the activity of NOS was significantly increased in 10 and 50mg/kg groups (P < 0.05), but not in 2mg/kg group (P > 0.05). There was no significant difference in the level of NO between the exposed groups and the control group (P > 0.05).

    Conclusion The intrauterine fenvalerate exposure exhibited obvious estrogenic effects on the male characteristics of offspring rats.
