Improving Effect of rhPTH on Hematopoietic Depression in Mice Caused by Chronic Benzene Poisoning

  • Objective To explore the improving effect of recombinant human parathyroid hormone(rhPTH) on bone marrow hematopoietic depression in mice caused by chronic benzene poisoning.

    Methods The model mice of hematopoietic depression was established in a number of KM mice by subcutaneous administration of benzene. Established model mice were treated 5 times a week for 3 weeks intraperitoneally with normal saline (NS group), 40 μg/kg rhPTH (40PTH group), and 80 μg/kg rhPTH (80PTH group) separately. One week later, all mice in the three groups were sacrificed. The hematopoietic stem cell, hematopoietic progenitor cell, peripheral blood cell count, and spleen index were determined by routine methods.

    Results Mice in PTH-treated groups showed better general condition than mice in NS group. The hematopoietic stem cells in 40PTH group and 80PTH group were significantly increased than those in NS group (P<0.05). However, the hematopoietic progenitor cells and peripheral blood counts had no significant differences among the three groups. The spleen index in 40PTH group and 80PTH group was notably lower than NS group (P<0.05).

    Conclusion rhPTH markedly increased the number of hematopoietic stem cells in mice with chronic benzene poisoning.
