Influence of Hypoxia on Islet Morphology of Streptozotocin-induced Diabetic Rats at High Altitude

  • Objective To discusses hypoxia influence on islet morphology of streptozotocin (STZ)-induced diabetic rats through observing islet morphology characteristics and measuring morphological index of Xi'ning and Muli diabetic rats.

    Methods Rats were randomly divided into Xi'ning normal group,Muli normal group,Xi'ning diabetic group and Muli diabetic group randomly.Rats of diabetic groups were intraperitoneally injected STZ after a 12-hour-fast.Animal models considered successful if blood sugar levels of rats were higher than 16.7 mmol/L.Islet area,beta cell area,light density,IOD and number of apoptotic islet cell were measured by immunohistochemical staining and TUNEL apoptosis test.

    Results The blood glucose levels of Xi'ning STZ rats were higher than that of the Muli STZ rats.The islet number of STZ rats reduced,islet atrophy with islet cell degeneration was also observed.The beta cells of the STZ rats reduced,insulin dyeing showed shallow.The apoptotic cells in the islets of STZ rats were more than the normal rats,and distributed uniformly.The Xi'ning diabetic rats and the Muli diabetic rats diabetes significantly different between the β cell area,the β cells area/islet size and the average IOD.

    Conclusion Islet hypoxia tolerance,faster glucose uptake and increased insulin sensitivity are likely explanations for STZ diabetic rats at high altitude showing lower blood sugar,minor damages to beta cells.
