The Protective Effect of Inonotus Obliquus Polysaccharide (IOP) on the Hepatic Injury Induced by Diethylnitrosamine

  • Objective To study the preventive effect of inonotus obliquus polysaccharide(IOP)on the hepatic injury induced by diethylnitrosamine(DEN).

    Methods Fifty mice were randomly divided into five groups:negative control group(Distilled water), DEN group(20 mg/kg), high dose of IOP group(600 mg/kg), middle dose of IOP group(300 mg/kg), and low dose of IOP group(150 mg/ kg). Mice of negative control group were given with distilled water and the three IOP groups were given with IOP, all by gavage daily for 5 weeks. Mice of DEN group and three IOP groups were given with DEN intraperitoneally every other day. After 5 weeks, the mice were killed, the number of liver cell mitosis and Kupffer cell were counted under microscope with HE staining. The activity of glutathione S-transferases (GSTs)and content of malondialdehyde(MDA)were determined by enzyme dynamic method and the content of tumor necrosis factor α (TNF-α)in liver tissue was determined by ELISA.

    Results The numbers of mitosis and Kuffer cell in DEN group were more than that of the negative control group with significant statistical difference(P<0.01), and the high dose of IOP group showed less mitosis and Kuffer cell than that of the DEN group with significant statistical difference(P<0.05). The activity of GSTs in DEN group was lower than that of the negative control group with significant statistical difference(P<0.05), the contents of MDA and TNF-α in DEN group were higher than that of the negative control group with significant statistical difference(P<0.01 or P<0.05). The activities of GSTs in the high dose and middle dose of IOP group were higher than that of the DEN group with significant statistical difference(P<0.01). The contents of MDA in three different dose of IOP groups were lower than that of DEN group with significant statistical difference(P<0.01 or P<0.05). Only in high dose of IOP group the content of TNF-α was significantly lower than that of DEN group with significant statistical difference(P<0.05).

    Conclusion IOP has some protective effect on the hepatic injury induced by DEN.
